La maladie d’Alzheimer a fait, au cours des dernières décennies, l’objet de travaux qui permettent de mieux comprendre, au niveau moléculaire, les causes de sa survenue. Le mécanisme essentiel semble impliquer la formation et l’activité de la β-amyloïde. La cascade d’événements impliqués dans sa synthèse à partir de son précurseur (l’APP) et sa dégradation a été, pour l’essentiel, identifiée. Cela débouche en théorie sur des possibilités thérapeutiques ciblant les causes mêmes de la pathologie. Cette recherche passionnante se heurte cependant à un obstacle théorique intéressant à savoir la méconnaissance de la fonction physiologique des molécules impliquées, notamment la β-amyloïde et son précurseur.
