The aim of the present polarization microscopic study was to ascertain whether there are differences between the modes of pharmacodynamic re- sponse of three test targets: malignant epidermal hyperplasia, benign epidermal hyperplasia, and normal epidermis in mice of the skin-tumor- resistant RA strain, to the effect of a threshold dosage of colchicine when a nonpolar-polar substance, dimethylsulfoxide (DMSO) acts as a carrier for the cytostaticum. Performance of the study was all the more so indicated as it was earlier demonstrated at this laboratory that there are, in fact, fundamental differences (statistically highly significant: P < 0.001) in the modes of response ofmalig- nant epidermal cells between series with colchicine in acetone and colchicine in DMSO. The differences manifested themselves at light- and electron microscopic levels, as well as when the relevant cell populations were ana- lyzed by means of the histoquantitative technique. Hence, and because earlier studies in this field had also revealed fundamental differences in the structure and functions of the keratin-stabilizing systems between the malignant and the benign epidermal cells, it could be expected in advance that certain occurrences may also develop at the macromolecular level revealable by the polarization microscopic technique.
