For many years metabolism of calcium and bone was investigated with metabolic balance and histologic techniques alone. More recently the advent of radioactive isotopes has made possible the determination of rates of calcium movement within the individual (1-3), Methods have been devised for the study of bone metabolism in vivo (4, 5). Urinary hydroxyproline has proved to be a useful index of collagen turnover (6-8). Studies with these procedures have greatly increased our understanding of normal processes as well as those involved in clinical diseases of bone. The ultimate goal, how ever, is to define clearly metabolic alterations at the cellular and molecular level and to correlate these changes with clinical aberrations. In this review will be presented an outline of the methods for kinetic studies of calcium metabolism in vivo, results of some of the studies with these methods, findings in some of the studies of bone metabolism in vitro, and a brief discussion of hydroxyproline metabolism. Because of its relevance to bone metabolism and because it is the subject of widespread current interest and intensive investigation, the most recent information concerning thyro calcitonin will also be presented.
